Combined effect of counting chamber and diluent on sperm kinematics

The evaluation of sperm motion is crucial for processing of seminal doses for artificial insemination.

The evaluation of sperm The evaluation of sperm motion is crucial for processing of seminal doses for artificial insemination. Here, the combined effect of the type and capture area of three counting chambers, together with the type of diluent employed, on sperm motility was analysed. Ejaculates from thirteen Holstein bulls were used for sperm kinematic analysis with the ISAS®v1 CASA‐Mot system, using two capillary‐loaded counting chambers (Leja® and Cell‐Vu®) and one drop displacement chamber (Makler®).

Spermiogram and sperm head morphometry assessed by multivariate cluster analysis results during adolescence

Spermiogram and sperm head morphometry assessed by multivariate cluster analysis results during adolescence (12-18 years) and the effect of varicocele.

Volunteers between 150 and 224 months of age, who had reached oigarche by 12 years old, were recruited in the area of Barranquilla, Colombia.

Genetic, environmental, and nutritional factors have a great influence on the time to reach puberty. Therefore, it is difficult to predict the age at which a particular individual will achieve his reproductive maturity, i.e., the sperm production corresponding to that of an adult.

In addition, biomedical information about the age of first conscious ejaculation (oigarche1), the presence or absence of spermatozoa in seminal fluid, the biological characteristics of spermatozoa during this maturational process, and the time required to reach adult spermatogenesis, is currently scarce and controversial.

The few studies reported on this topic correspond to adolescents under the age of 18 years and are related to different pathologies such as testicular cancer.

The main objective of this study was to investigate sperm features in adolescents between 12 and 18 years of age. These data are part of a social project carried out in Barranquilla, Colombia, directed to develop a sexual educational program because of the high teenage pregnancy rate in this area. Therefore, it is necessary to know the mean age at the onset of sperm production in adolescents from this area to provide the best counseling.

In addition, we analyzed the effect of varicocele and used morphometric data analysis to assess sperm subpopulations in human semen samples from these adolescents.

Disposable chambers for cell counting . An option for your counting.

Disposable chambers for cell counting . An option for your counting.

A simple option to help in the laboratory when performing the analyzes. There are a diversity of designs and models that conform toth especifications of our customers.

Choose between D2C and D4C models, the difference is between the number of samples per camera. In both models you have to choose different depths:

  • 10 µm: The “clasic” for human semen analysis (WHO 1999).
  • 16 µm: Used in many farm seminal analysis.
  • 20 µm: Currently recommended for human semen análisis by WHO 2010. Optimal for veterinary.

Disposable chambers are indicated for concentration estimate and analysis of motility with CASA system. Pre-drawn squares assure an optimal sampling.

Disposable chambers

One box have 25 slides for 50 or 100 analysis, all depend the model for disposable chambers.

How sexual competition can affect the function and quality of sperm

The presence of a rival male influences the quality of sperm, according to research led by the University of Alicante (UA) and the University of Upsala (Sweden).

The presence of a rival male influences the quality of sperm, according to research led by the University of Alicante (UA) and the University of Upsala (Sweden). This work was done with adult zebra fish (Daniorerio), reveals that the spermatozoa of males exposed to high competition (two males and one female) present changes, resulting in faster and more competitive sperm. Anyway, this is not a good way of acting, since it leads to a change in the next generation with respect to DNA integrity.

The methodology followed in the investigation consisted of placing two males for two weeks with one female and two females with one male, this would give a treatment of high competitive ness and low one. The difference in both treatments was observed and a possible explanation that brings negative effects on DNA integrity is that due to the competitive ness generated in the environment there is a high production of reactive oxygen species caused by stress.

That is to say, the quality of the spermatozoa of a seminal sample does not depend on the quality of them individually, but rather on the joint action of diverse sperm subpopulations and also depends on the joint action of the individuals of the species and their sexual-competitive relationships.

The work was published in the scientific journal Journal of Evolutionary Biology.

evaluate sperm morphology

Evaluate sperm morphology

How do you do stain to evaluate sperm morphology?

Approximately 15% of couples have infertility problems. Evaluate sperm morphology is an important parameter forsuccessful fertilization. In assisted reproduction techniques (Kuster et al., 2004) the Kruger criteria(1986) and the WHO classification are the most important. According to the Kruger criteria, the head, neck and tail of the sperm must be normal and 70% of the head must be covered by the acrosome.

Sperm quality and behavior of subpopulations.

Sperm quality and subpopulations behaviour.

The quality of the sperm from a semen sample does not depend, as was previously thought, on the individual quality of the semen, this depend the joint action of many sperm subpopulations.

espermatozoide
This conclusion, which is based on research carried out in recent decades by various research groups, has been corroborated by the joint work of researchers from the University of Valencia and the PROISER R + D, S.L.

The study on the sperm behaviour and their quality in samples of diverse species has been one of the main objectives of the research group by Dr. Carles Soler in the Departament of Celular biology, Functional Biology and Physical Anthropology of the Universitat de València for several decades. A line of research that has been linked to the approaches of I D I from the Valencian company PROISER, dedicated to the development of solutions for the diagnosis of semen samples. They have developed a revolutionary software to study the divers quantitative parameters of sperm samples.

As a result of the experience acquired, Dr. Carles Soler, as head of the University’s research group and as scientific director of PROISER, was invited to be the editor of two special issues in the prestigious journals Asian Journal of Andrology (2016) and Reproduction, Fertility and Development (2018), respectively, with morphometry and sperm kinetics.

The CASA systems (Computer Assisted Semen Analysis) developed by PROISER provides a databases for tens of thousands of sperm. The application of advanced statistical techniques of multivariate analysis (main components and cluster) from these databases have allowed to consolidate the concept of subpopulations.

Hitherto, has been thought that the behavior of sperm within an ejaculate was comparable to what happens in a marathon: many runners, in principle equivalent, competing for the goal that is the oocyte. Now, the verification mathematically coherent subpopulations points more or less organized team concept.

All of this is around about what happen the sperm in a female tract. Each spermatozoonfind many obstacules and barriers that help to select the best sperm. We can say that it is similar to chess. It would not make sense to spend the towers or even the king in the opening and the first dabbling, that’s what the pawns are for.

espermatozoideIn any case, we still do not know what the rules that the sperm through in the female genital tract. The essay by the Universitat de València and PROISER, are currently work for to find more solutions.

Referent in the reproduction area

In addition to the physiological conditions, these results should lead to the definition of changes in the strategies followed in assisted reproduction, both in their veterinary and human applications. This group of studies and the new perspectives that have consider turned the group of the Universitat de València and PROISER into world leaders.

This new points come in addition to other PROISER findings in the same area. It is this private company has been developing software and computer applications for more than a decade that help to do a successful seminal analysis.

In this way, PROISER continues to advance and placing Valencian talent at the top of the international ranking.
